New users

Each group leader needs to register here. BREM will provide credentials for booking through Agendo management software. A brief user guide can be found here.

Remote access to our microscopes requires a TeamViewer account. 

Familiarize with the Resources, Procedures, Guides and Policies of BREM.

Before a session

A session can be scheduled through Agendo. Users may sign up for up to one 24 hour session from Monday to Wednesday. The acquisition rate on a standard single particle data collection session is >750 movies per hour.

Users should ship vitrified grids to BREM in advance of their agreed data collection session. If you ship unclipped grids, they are due at BREM no later than Wednesday @ 14:00 CET the week before the scheduled session. Pre-clipped grids are due at BREM no later than Thursday @ 14:00 CET the week before your scheduled session. If the grids don’t arrive in due time the Krios G4 data collection session will be cancelled and rescheduled. At BREM we accept grids up to 3 weeks ahead of your session.

We encourage users to operate the Krios G4 remotely alongside our staff. To this end, log-in to your TeamViewer account. No hand panels or additional hardware are required besides a large (>20”) LCD screen.

During a session

A maximum of up to 3 clipped grids are loaded onto the Krios G4 per 24 hour data collection session. A typical schedule for a Single Particle or Tomography 24-hour data collection session is as follows:

·  9:00-10:00: On Mondays clipped grids for the Monday-Tuesday-Wednesday sessions are loaded onto the Krios G4 by BREM staff

·  10:00-11:00: The GIF is tuned and a new K3 gain reference is acquired by BREM staff

·  11:00-13:00: User remotely selects targets for data collection

·  13:00-14:30: EPU or Serial EM is setup on suitable locations

·  14:30-17:00: Data collection begins while is monitored and adjusted

·  17:00-9:00 next day: Data collection proceeds unsupervised 

Movies will be motion corrected and CTF estimated on-the-fly, and initial particle picking and 2D classification will be carried out automatically in order for users to evaluate the data collection session. 

After a session

User should arrange pick-up and return shipping of dewars.

Users should copy the data from a session through bandwidth file transfer systems.

Data from a session will be queued for deletion after 30 days.

BREM must be informed about all publications derived from the use of its instrumentation, and BREM itself and relevant BREM funding sources must be acknowledged.
